ความหลากหลายทางพันธุกรรมของสายพันธุ์ข้าวพื้นเมืองในภาคใต้ของไทยจากลักษณะทางสัณฐานวิทยาของเมล็ดข้าว Thai Agricultural

Seed morphology of 50 local rice varieties in the southern Thailand was studied in length, width, 100 seed weight, colour, and hair of husked seed and length, width, length/width ratio, shape and colour of dehusked seed. Five rice varieties of central Thailand were used as an outgroup. There is variation in all characters measured except the hair of husked seed and colour of dehusked seed. Husked seed width and dehusked seed width and length/width ratio have normal distribution. Dendrogram showing the relationship among rice varieties will be presented in the meeting.

Variation in zinc (Zn) concentration in the grain of 50 local rice varieties from Southern Thailand was examined by diphenyl thiocarbazone (DTZ) staining (intense, medium, low, and nil) and chemical analysis. Intense staining was found in the aleurone layer of 94% of the varieties, in the embryo 56% of the varieties and endosperm 12% of the varieties. Variation was found in the DTZ staining intensity of the embryo and endosperm of individual grains, with Shannon-weaver diversity index (H) of the DTZ staining at 0.15-1.00 for the embryo and 0.00-1.00 for the endosperm. Crude protein extracts from Thai rice seeds were screened for α-amylase inhibitory activity. At least 5 rice seed extracts exhibited a potent α-amylase inhibitory activity with specific activity above 50% inhibition per μg protein. Those α-amylase inhibitory activities have no positive correlation with α-amylase activity in each rice sample. Denatured SDS-PAGE revealed major proteins bands with molecular mass around <20, 28 and 40 kDa. Anion-exchange column chromatographic profile of Q-Sepharose XL at buffer system of 8.5 showed the bound proteins contained α-amylase inhibitory activity. Paddy storage may cause considerable losses and changes in quality of rice. The objective of this study is to determine changes in some physical and chemical properties of rice grains during storage. The experiment was conducted at Pathumthani Rice Research Center during May 2012 – January 2013. PTT1 variety was used for low amylose rice, and PSL2 for high amylose. Samples were taken and stored in 2 kg woven plastic sack at room temperature. The PTT1 and PSL2 samples were stored for 8 and 6 months, respectively.
